Lateral flow immunoassay using europium chelate-loaded silica nanoparticles as labels.
BACKGROUND Despite their ease of use, lateral flow immunoassays (LFIAs) often suffer from poor quantitative discrimination and low analytical sensitivity. We explored the use of a novel class of europium chelate-loaded silica nanoparticles as labels to overcome these limitations. METHODS Antibodies were covalently conjugated onto europium chelate-loaded silica nanoparticles with dextran as a ...
متن کاملEnhanced lateral flow immunoassay using gold nanoparticles loaded with enzymes.
The use of gold nanoparticles (AuNPs) as labeling carriers in combination with the enzymatic activity of the horseradish peroxidase (HRP) in order to achieve an improved optical lateral flow immunoassay (LFIA) performance is presented here. Briefly in a LFIA with an immune-sandwich format AuNPs are functionalized with a detection antibody already modified with HRP, obtaining an 'enhanced' label...
متن کاملTime-Resolved Luminescence Microscopy and Microarray Using Europium Chelate Labels
The luminescent Eu complex, DTBTA-Eu, is highly stable in terms of ligand-metal dissociation, and can be labelled to amino groups of proteins and other bio-molecules. Owing to the long lifetime of Eu luminescence, time-resolved measurement of DTBTA-Eu removes background fluorescence and enhances the sensitivity of the labelled materials. This property was applied to time-resolved luminescence m...
متن کاملLateral flow immunoassay using Europium (III) chelate microparticles and time-resolved fluorescence for eosinophils and neutrophils in whole blood.
BACKGROUND A simple point-of-care method for measuring leukocyte counts in a doctor's office or emergency room could be of great importance. We developed a protocol for measuring cell count by disrupting the cell membrane and analyzing specific proteins within the cells and used it to analyze proteins from eosinophils and neutrophils. METHODS Lateral immunochromatographic (ICR) assays have be...
متن کاملDye-doped silica nanoparticles as efficient labels for glycans.
We report that dye-doped fluorescent silica nanoparticles (FSNPs) are highly efficient labels for glycans. Mono- and oligo-saccharides were conjugated to FSNPs using a general photocoupling chemistry. FSNP-labeled glycans were applied to image and detect bacteria, and to study carbohydrate-lectin interactions on a lectin microarray.
